Following up on the Sunday evening workshop at ERCC9, “Vesicle Isolation & Function: Translating vesicle biology to clinical therapy,” the goal of this mini-conference is to further discussion on optimal separation strategies for functional testing of extracellular vesicles (EVs). Our goal is to identify the most promising separation strategy or strategies for scaling up vesicle yield while maintaining functional activity of vesicles. We will compare the merits of conventional ultracentrifugation (UC), UC followed by density gradient, tangential flow filtration (TFF), and size exclusion chromatography followed by TFF, and work towards developing a joint program to push that strategy towards the manufacturing phase.
